CAR-T Cell Therapy Resistance Modeling: Investigating Allee and Bystander Effect

Recent research on CAR-T cell immunotherapy has focused on identifying and addressing the mechanisms behind patient relapses following treatment. Among the observed dynamics, the Allee and bystander effects have emerged as significant factors influencing tumor growth, stabilization, and elimination. To investigate how these effects impact therapy outcomes, we developed a mathematical model using a system of differential equations. This model incorporates heterogeneity in the expression of the target antigen among tumor cells, enabling the classification of tumor subpopulations as either sensitive or resistant to therapy. Through simulations exploring varying strengths of these effects and different proportions of resistant tumor populations, we analyzed their influence on disease progression, providing insights into the complex interplay between tumor dynamics and therapeutic efficacy.
